There are several approaches to investigate this:

Home urine tests are available however, they only will identify drug use in the last 2-3 days in most cases and can be tampered with by diluting the specimen with water or other chemicals.

Hair testing - provides a longer period of detection, however lighter/intermittent use may go undetected.

Physician evaluation - I would actually suggest starting with a visit to the GP. There are a number of clinical findings on physical examination that may identify or narrow the list of possible drugs that could be involved. The GP can also help you in obtaining the right type of testing and discussing treatment options if he is indeed using drugs.
