Is it an offence to accuse somebody of something they have not done ?
Like if your wife accuses you of sleeping with another women when you have not.
Is that an offence ?

It is not a criminal offence, no.
What you are talking about is one private citizen saying something untrue about another private citizen. That is not a criminal offence but could be defamatory. Defamation is a civil matter which could result in the aggrieved person raising civil proceedings for damages.
