ISA's do not come into the equation on establishing whether your wife needs to send a tax return or not.

Not everyone has to file a tax return. If HMRC send a notice to your wife that she needs to file a tax return then she has no choice but to file one.
If your wife's income is below the threshold for higher rate tax and all income is taxed at source, then it is most likely that she would not have to send in a tax return.
