First, try and restart the printer and the computer. Sometimes that fixes this issue.

Good. Now open a browser and go to https://s3.amazonaws.com/download.dymo.com/dymo/Software/Win/DCDSetup1.4.3.131.exe
Install the download and it will walk you through adding it back.
When installing the driver?
Try plugging up the USB and see if it reinstalls it.
Right click and choose test print.
